| I. Background | |
| CD47 是一種廣泛表達(dá)于多種細(xì)胞表面的跨膜糖蛋白基因，屬于免疫球蛋白超家族成 員。其編碼的蛋白質(zhì)在細(xì)胞表面發(fā)揮重要功能，主要通過(guò)與信號(hào)調(diào)節(jié)蛋白α（SIRPα）結(jié)合， 傳遞“別吃我”信號(hào)，抑制巨噬細(xì)胞等免疫細(xì)胞對(duì)自身細(xì)胞的吞噬，從而在免疫逃逸和自 身耐受中起關(guān)鍵作用。在腫瘤微環(huán)境中，癌細(xì)胞常高表達(dá) CD47，借此逃避免疫監(jiān)視，促 進(jìn)腫瘤生長(zhǎng)與轉(zhuǎn)移。此外，CD47 還參與細(xì)胞遷移、增殖及凋亡等過(guò)程，并與血小板活化、 神經(jīng)發(fā)育等功能相關(guān)。研究表明，CD47 在多種血液系統(tǒng)惡性腫瘤及實(shí)體瘤中高表達(dá)，與 不良預(yù)后相關(guān)，使其成為腫瘤免疫治療的潛在靶點(diǎn)。目前，針對(duì) CD47 的阻斷抗體或融合 蛋白等療法正在臨床研究中，旨在恢復(fù)巨噬細(xì)胞對(duì)腫瘤細(xì)胞的吞噬作用，為癌癥治療提供 新策略。 | |
