ADCP(Antibody-dependent cellular phagocytosis,抗體依賴性細(xì)胞介導(dǎo)的吞噬作用)是一種重要的免疫清除機(jī)制。治療性單克隆抗體(mAb)結(jié)合靶細(xì)胞后,其Fc段可被吞噬免疫細(xì)胞(如巨噬細(xì)胞、單核細(xì)胞、中性粒細(xì)胞)表面的Fcγ受體(FcγR)識(shí)別,從而介導(dǎo)對(duì)靶細(xì)胞的吞噬清除。 在人類(lèi)中,F(xiàn)cγR家族包含激活型受體(如FcγRIIa/CD32a、FcγRI/CD64、FcγRIIIa/CD16a)和抑制型受體FcγRIIb(CD32b)。FcγRIIb是唯一表達(dá)于免疫細(xì)胞表面的抑制性Fcγ受體,其胞內(nèi)段含有一個(gè)免疫受體酪氨酸抑制基序(ITIM)。當(dāng)FcγRIIb與抗體Fc段結(jié)合并被交聯(lián)時(shí),ITIM被磷酸化,招募含SH2結(jié)構(gòu)域的肌醇磷酸酶(SHIP),進(jìn)而抑制下游激活信號(hào)通路,從而負(fù)向調(diào)控ADCP及其他效應(yīng)功能。FcγRIIb在維持免疫穩(wěn)態(tài)、防止過(guò)度炎癥反應(yīng)中發(fā)揮關(guān)鍵作用,同時(shí)也是腫瘤免疫治療和抗體藥物研發(fā)中重要的潛在靶點(diǎn)。 |
