| I. Background | |
| 跨膜激活劑和 CAML 相互作用子（TACI）是腫瘤壞死因子受體（TNFR）超家族的成 員，也被稱為 TNFRSF13B。TACI 是免疫反應的調節(jié)因子，可抑制 B 細胞擴增，促進漿細 胞分化和存活。 TACI 配體是 BAFF 和 APRIL。BAFF 是屬于 TNF 配體超家族的 II 型跨膜蛋白，以膜 結合形式和可溶形式的三聚體兩種形式存在。APRIL 屬于配體 TNF 超家族，是 II 型跨膜蛋 白，參與晚期 B 細胞分化、抗體分泌漿細胞的產(chǎn)生和維持以及 IgA 類轉換。髓系細胞（包 括巨噬細胞、樹突狀細胞和血液單核細胞）以及多形核細胞（如中性粒細胞、嗜酸性粒細 胞和黏膜上皮細胞）負責 APRIL 的產(chǎn)生。APRIL 對 BCMA 表現(xiàn)出強結合親和力，對 TACI 具有中等親和力，而 BAFF 與 BCMA 的結合較弱，與 TACI 具有較強的親和力。TACI 與 APRIL 和硫酸乙酰肝素蛋白聚糖 （HSPG） 相互作用，促進 TACI 高效信號傳導以發(fā)揮其 生理效應。 |
